This work is devoted to the modification of the method for determining the activity of both the whole cellulolytic complex of all enzymes, and the activity of individual enzymes of this complex, namely, hydrolytic and monooxygenase cellulases. The aim of the work was to determine the effect of coloring substances on the determination of known glucose concentrations, by the method of determining the construction of a calibration curve. Modification of the method involves the introduction of sulfuric acid to precipitate humic substances and the exclusion of the dilution stage with distilled water at the temperature control stage before determining the optical density by spectrophotometric method. The assessment of cellulase activity in natural soddy-podzolic sandy loamy soil was studied. As a result of the research work, it was found that the cellulase activity determined by the standard and modified method during soil testing and the addition of gallic acid with the precipitation of sulfuric acid differ significantly by almost 12 times, with the addition of cellulose, the activity increases from 0.16% to 9.89%, i.e. increases by almost 62%.
